Synthesis of Low Molecular Water Soluble Chitosan Conjugated Biotin for Utilizing Target Drug Delivery System

Abstract

|
The aim of this study is to prepare delivery vehicles of methotrexate using Low molecular water soluble chitosan (LMWSC) and investigate them as an anticancer target drug delivery system. LMWSC was conjugated with methotrexate (MTX), and it was then modified with biotin and methoxy polyethylene glycol(mPEG) (biotin-PEG-LMWSC-MTX, BPLMMTX). BPLMMTX were prepared by the dialysis method, and the structure was confirmed by furier transform infrared(FT-IR) and 1H nuclear magnetic resonance (1H NMR) analysis. BPLMMTX has around 100-200 nm in particle size and observed to spherical shape by transmission electron microscopy (TEM). In addition, the cytotoxicity and antitumor activity of BPLMMTX were evaluated against L929 and, MCF-7 and KB cell lines by using MTT assay, respectively.
